Development and validation of ultra-fast quantitative real-time PCR method to differentiate between Oncorhynchus keta and Oncorhynchus mykiss

The ultra-fast quantitative real-time polymerase chain reaction (qPCR) assay was developed and validated to differentiate the morphologically similar ones, Oncorhynchus keta mykiss. Species-specific primers were designed for COI genes of mtDNA. species-specific O. mykiss selectively amplified by DNA, respectively. sensitivity 1 ng/μL. Quantitative testing showed that results met ‘Guidelines on Standard Procedures Preparing Analysis Method such as Food’ proposed Ministry Food Drug Safety. qPCR method in this study identifying has advantages speed field applicability. Therefore, is expected help control forgery alteration raw materials seafood industry.
